How are leading research institutes addressing the urgent need for novel TB therapies? In our tutorial led by Peter Craggs, Pharmacology Expert at The Francis Crick Institute, we learned how a multi-partner collaboration is building on previous GSK efforts to identify and validate new targets against an under-explored area in TB drug discovery.
In this work, a high-throughput repurposing screening approach has been taken to identify inhibitors from a library of 6,000 existing compounds. This project was enabled by building an integrated automated workflow in collaboration with SPT Labtech and Automata. The non-contact positive displacement dispenser, dragonfly® discovery from SPT Labtech, delivered fast and precise liquid handling, while the Automata LINQ platform allowed seamless end-to-end assay automation of such a time-sensitive kinetic assay. This collaborative, integrated approach accelerated the discovery of novel Mtb enzyme inhibitors and laid the groundwork for further medicinal chemistry efforts to develop this promising class of antitubercular agents.

We demonstrate that the Roche KAPA EvoPlus V2 and KAPA EvoPlus V2 PCR-free DNA library preparation workflows can be successfully automated on firefly to create Illumina-compatible NGS libraries comparable to those prepared manually. The combination of these technologies enables significant time savings in the laboratory for whole genome sequencing applications, whilst also reducing operator variability.

This poster highlights how mosquito HV genomics has been used to automate the Illumina Nextera XT protocol at SEQ-IT, achieving 5X miniaturized reaction volumes while delivering reproducible, high-quality libraries. This miniaturized method was developed to identify and screen mutations with oncogenic potential and drug resistance (HIV, HBV, HCV), SNP detection, as well as in HLA-typing, among other applications.

The ICESTP Safety Panel 90 includes 138 assays, incorporating both single-point screening and dose-response screening with a functional assay format, such as Automatic patch clamp, FLIPR calcium flow, and HTRF, for example. In this poster, we demonstrate the use of firefly® for high-throughput sample processing to facilitate the safety assessment of a 90-target panel, to improve the efficiency and accuracy of drug safety evaluations.

The DToL initiative aims to advance our understanding of life on Earth by unravelling the genetic blueprintt of a wide range of organisms. To address the scale of this challenge, the team at the WSI describe the development and validation of a novel automated method on firefly® to construct whole genome sequencing libraries from genomic DNA using the PacBio SMRTbell prep kit 3.0.

Cellular agriculture could be instrumental to environmentally-friendly food production. However, high raw materials costs impede financial viability. This poster highlights how the team at Uncommon have applied automated liquid handling and Design of Experiment (DoE) methodology to drive the development of low-cost, animal-free media.

In this study, we explore the benefits of automating the Scale Bio single cell methylation protocol with SPT Labtech’s firefly® to facilitate the analysis of single cell methylomes isolated from human peripheral blood mononuclear cells (PBMCs) compared to a library prepared in parallel using manual methods.

We demonstrate the ease of automating NEB’s UltraExpress® kits with the user-friendly firefly system, achieving high-quality and reproducible libraries and sequencing data. Libraries were prepared from 25 ng to 250 ng of total RNA, consistently showing high data quality across this input range.
